The OED says it's short for Gräfenberg spot: "a sensitive area of the anterior wall of the vagina believed by some to be highly erogenous and capable of ejaculation (first described by Gräfenberg & Dickinson 1944, in Western Jrnl. Surg. 52 338); abbrev. G-spot." (VSD)
