it has a sarcastic meaning of "i doubt that" or "is not so", expressing doubt towards the subject expressed.

it is often used as a singular reply to another person who may be "pulling your leg".

not sure of origin, but I remember it as far back as ~early 60's (US).

Of course, Mr. O'Toole meant it in gracious humor, refering to his missing winning the award in earlier years. IMO, his speech was the most poignant moment in this year's show.
